Unlike the first game, Elite Force II is largely set onboard the USS Enterprise-E, stationed in the Alpha Quadrant. The game's storyline is a semi-sequel to the movie Star Trek: Nemesis, and the end of Star Trek: Voyager series. Many of the members of the Hazard Team from the first game reappear. However, unlike in the first game, only a few of the primary cast of Star Trek: The Next Generation provided voice acting for this game. Appearing cast members include Patrick Stewart as Jean-Luc Picard, Dwight Schultz as Reginald Barclay, and Tim Russ as Tuvok (brought over to the Enterprise-E from Voyager). There are several guest star actors from several Star Trek Series who do some excellent voice acting like, Jeffrey Combs, who previously portrayed Weyoun on Star Trek: Deep Space Nine and Commander Shran on Star Trek: Enterprise, provides the voice of one of the game's main villains. Additionally, Robert O'Reilly (who was Gowron in Deep Space Nine) provides the voice of Krindo, and Vaughn Armstrong (Admiral Forrest in Enterprise) provides several character voices, Lastly the great J.G Hertzler (Martok in Deep Space Nine, plus roles in Voyager and Enterprise)provides the voice of Lurok.
