i've always been online - when activating the license.
so i'd say that you can't - if there is no 'net to that machine . . .
I think activation is a one time even, but you need to be internet connected for it happen. After that I think you can have it safely unconnected.

The other responses are correct: activation is a one-time event, but you must be connected to the internet during activation.
If you absolutely must activate on a machine which is not connected, please file a ticket with customer support. Mention in the ticket that you are simply unable to connect the machine in question to the Net, and they will be able to help you.
(Obviously, an unconnected version will not be able to receive things like tie file updates from our server.)
