The only issue you may have with using PyLotRO with Lord of the Rings Online is getting a suitable version of patchclient.dll, this only really applies to people using the original Shadow over Angmar CD/DVDs as the current digital downloads all contain a suitable version of patchclient.dll.
Configuration is easy, use the configuration wizard select Lord of the Rings Online (or LotRO Test - there are two "slots" so you can have two versions of the game, for example normal and the test server version) and your game directory should be found.
